Inhibitory Effects of Phenolic Portion from Kale Extract on N-Nitrosodimethylamine Formation

Abstract
Kale extract was separated into phenolic and ascorbate portions using a sep-pak C_18 cartridge. The phenolic portion was further fractionated using HPLC and prep-LC. These phenolic fractions (fractions 1, 2, 3 and 4) inhibited N-nitrosodimethylamine (NDMA) formation by 5.5 to 55.5% in the reaction mixture of 3.0 mL 0.15 M NaNO_2 and 1.5 mL 0.3 M dimethylamine. The fraction number 2 was further fractioned into 3 subfractions (subfractions 2a, 2b and 2c) using HPLC. The subfraction 2b especially inhibited NDMA formation by up to 80.4%. One of these fractions was a phenolic compound containing 25 carbons according to the analysis done with NMR and IR.
